The question
How do different intravenous doses of DMT behave in the body, and what acute effects do they produce in healthy people? Does the way doses are given, blinded or openly escalating, change the experience?
What they did
In one arm, 20 healthy participants received placebo and four doses of DMT (5, 10, 15 and 20 mg) as a rapid injection, in random order, within a single test session, with neither participants nor staff knowing which was which. In a second arm, 16 participants knowingly received stepwise 5 mg increases until they reached the highest dose they could tolerate, up to 25 mg. Researchers tracked subjective effects, heart and circulation measures, side effects, and blood levels of DMT for up to 55 minutes after each injection.
What they found
Effects came on very quickly, peaking within the first 2 minutes, then faded and were completely gone within 12 to 30 minutes. This matched a short elimination half-life of about 6 to 7 minutes, meaning the drug left the blood rapidly. Peak subjective effects levelled off at the 15 mg dose, so higher doses did not feel stronger. Repeated dosing in one session did not produce tolerance. Tolerability was clearly better with open-label dose escalation than with blinded random dosing, and at the same dose levels participants rated the effects as less intense when they knew what was coming.
The fine print
This was a small study in healthy volunteers, not people with a mental health condition, so it says nothing about whether DMT helps depression. Follow-up was limited to under an hour after each dose. The dose-escalation arm was open-label, so expectancy could shape both ratings and tolerability. This summary is based on the abstract alone.
What it means
The study maps out how quickly intravenous DMT acts and clears, and suggests that above about 15 mg the peak experience does not intensify. It also shows that knowing what dose you are getting can change how intense the experience feels, which matters for how future trials are designed.
Evidence grade: Moderate. A randomized, placebo-controlled crossover trial, but small, in healthy volunteers only, with a separate non-blinded arm and no clinical outcomes.
